Heck \u2013 right now, somewhere on your body, you\u2019ve probably even got an itch caused by nothing in particular. Well, scientists at Harvard Medical School in the USA have discovered its cause: <a href=\"http:\/\/10.1016\/j.cell.2023.10.019\" target=\"_blank\" rel=\"noreferrer noopener\">a (very gross) bacteria living in your skin<\/a>.<\/p><p>And (as if it couldn\u2019t get worse) the researchers have found this happens because the bacteria, known as <em>Staphylococcus aureus<\/em>, literally changes your nerve cells. If your skin wasn\u2019t crawling before, we bet it is now.<\/p><p>But how does it make you itch? Published in the journal <em>Cell<\/em>, a new paper reveals that the pesky little microbe releases a chemical which activates a protein in our nerves. This sends a signal from our skin to our brains \u2013 which our brain perceives as an itch.<\/p><p>If it makes you feel any better, there are plenty of <em>good<\/em> bacteria that also live in your skin. In fact, <a href=\"https:\/\/www.sciencefocus.com\/the-human-body\/getting-under-your-skin-the-surprising-life-of-our-largest-organ\">this amazing organ<\/a> is kept healthy by a fine balance of microorganisms that call your skin their home.<\/p><p>But it\u2019s when this finely balanced <a href=\"https:\/\/www.sciencefocus.com\/the-human-body\/human-microbiome\/\">microbiome<\/a> becomes out of whack that <em>S. aureus<\/em> can go rampant.<\/p><p>The researchers discovered this through research on mice and human cells. To start, they exposed the skin of mice to <em>S. aureus<\/em>. The mice developed increasingly intense itches \u2013 directly due to the bacteria, but also in response to stimuli that shouldn\u2019t be so irritating, like a light touch.<\/p><figure class=\"wp-block-embed is-type-video is-provider-youtube wp-block-embed-youtube wp-embed-aspect-16-9 wp-has-aspect-ratio\"><div class=\"wp-block-embed__wrapper\"><iframe loading=\"lazy\" title=\"Scientists finally work out what causes itching - and it\u2019s grosser than you think\" width=\"800\" height=\"450\" src=\"https:\/\/www.youtube.com\/embed\/zbBQtb7LgLQ?feature=oembed\" frameborder=\"0\" allow=\"accelerometer; autoplay; clipboard-write; encrypted-media; gyroscope; picture-in-picture; web-share\" allowfullscreen=\"\"\/><\/div><\/figure><p>Next, the scientists isolated a single enzyme that they knew the microbe releases upon skin contact. They tested just this enzyme on the mice and measured levels of this enzyme in different human patients\u2019 skin. The scientists&#8217; conclusion: this enzyme is the single-handed culprit behind the itch.<\/p><p>In fact, patients with dermatitis have higher levels of this enzyme, known as protease V8, than people with healthy skin.<\/p><p>Proteaste V8 activates a protein in our nerves called PAR1, which usually lies dormant but awakens when the V8 \u2018snips\u2019 one end of it. The researchers tested V8 in lab dishes containing human neurons, and found these also responded to the enzyme.<\/p><h2 id=\"h-scratching-the-surface-of-itch-research\">Scratching the surface of itch research<\/h2><p>The discovery explains why people with eczema and atopic dermatitis itch a lot.<\/p><p>If you\u2019ve had a bad itch, you know how good it feels to scratch it \u2013 but you\u2019ll probably also know that it usually makes things worse. Itching, especially for those with skin conditions, can actually cause skin damage and even <a href=\"navto:\/\/ceb90f8a-1df5-4df1-8ca8-c70f4754f8d9\">pain<\/a>, the researchers warn.<\/p><p>That\u2019s why they\u2019re hoping that their findings will improve medicines and creams designed to treat itching linked to imbalances in the skin microbiome (like dermatitis and psoriasis).<\/p><p>How? The researchers knew that PAR1 (the protein that the microbes activate) is involved in blood-clotting, so they tried an approved anticlotting drug to treat the itching \u2013 and it worked. The rate of itching and resulting skin damage lowered in the mice.<\/p><p>What remains unclear is why the bacteria would <em>want<\/em> to make our skin crawl, evolutionarily speaking.<\/p><p>\u201cIt\u2019s a speculation at this point, but the itch-scratch cycle could benefit the microbes and enable their spread to distant body sites and to uninfected hosts,\u201d said first author <a href=\"https:\/\/chiulab.med.harvard.edu\/people\/liwen-deng\" target=\"_blank\" rel=\"noreferrer noopener\">Dr Liwen Deng<\/a>.<\/p><p>\u201cWhy do we itch and scratch?
